It depends on what your qualifications are. Do you mean largest in number of chapters and colonies, or do you mean number of initiated members, or both?

I was IFC Recruitment Coordinator two years ago and there was much chest thumping regarding sizes. I was putting together the rush guide for prospects so instead of relying on the chapters for numbers, I called EVERYONES headquarters. Some were cool, some were dicks. I don't remember the lower and latter numbers, but I do remember the top three in both categories.

Number of Chapters and Colonies:
1. Tau Kappa Epsilon - 305
2. Sigma Phi Epsilon - 265
3. Kappa Sigma - 245

Number of Initiated Members:
1. Sigma Alpha Epsilon - 255,000
2. Lambda Chi Alpha - 230,000
3. Sigma Chi - 220,000

Size doesn't always mean strength. Some on the list are relatively young. Others not on list have large financial reserves. Another interesting statistical breakdow is national average chapter size. Most likely only headquarters staff would know (or care to) this info, but one chapter on our campus was very large nationally, but had a national average chapter size of 14 men - (many chapters, but not many really large ones) While another chapter on my campus claimed to be "really selective about where it colonized," but had many initiates. Its average national chapter size was 54 men.

Boring I know, but you wanted stats.
That's what rush chairs do http://www.greekchat.com/forums/ubb/smile.gif

For women's fraternities, I know that Chi Omega, Delta Delta Delta, Delta Zeta and Kappa Kappa Gamma are probably the largest 4 in terms of total no. of initiates.
